The FITC / PE Compensation Standard is to be used in conjunction with hardware or software to remove spectral overlap from fluorochromes into secondary fluorescence detectors of a flow cytometer. The compensation can be done through different flow cytometry software such as Flowjo, Flowlogic, Kaluza etc. In cytometry, compensation is a mathematical correction of a signal overlap between the channels of the emission spectra of different fluorochromes. This ensures that the fluorescence detected in a particular detector derives from the fluorochrome that is being measured. To correct for this spectral overlap, a process of fluorescence compensation is used. However, when emission spectra overlap, fluorescence from more than one fluorochrome may be detected. Why is compensation of fluorescence necessary? Why is compensation important in flow cytometry?īecause the fluorophores used in flow cytometry emit photons of multiple energies and wavelengths, a mathematical method called compensation was developed to address the measurement of the photons of one fluorophore in multiple detectors. Special care must be taken to ensure that the compensation beads you choose will bind to the species in which your fluorochrome- conjugated antibody was raised. Compensation beads are small particles (typically polystyrene) that are pre-coated with antibodies recognizing species-specific antibody light chains.
